Description

The chronic degenerative disease osteoarthritis affects around 60% of people worldwide and is characterized by a progressive degradation of the cartilage. Patients suffer from pain, invalidity and dramatic loss of life quality. Besides medicamentous treatments, transplantation of autologous chondrocytes, the only cells within the cartilage, or total knee replacement by an implant are the only therapies to improve the clinical picture of osteoarthritis. A new regenerative approach is the use of patient´s blood derivatives. Platelet-rich plasma (PRP) and hypACTTM serum are the two most prominent among these. These products are an enrichment of various factors (especially growth factors) which accelerate the healing process of cartilage defects. OrthoSera GmbH has developed the CE marked device hypACTTM inject. With this syringe blood from the patient can be retrieved, centrifuged and processed within the same medical procedure. The obtained hypACTTM serum within hypACTTM inject can then directly be injected into the defective side. Together with exosomes and apoptotic bodies, microvesicles form the group of the so called extracellular vesicles which represent a novel mechanism by which pathogenic signals are communicated among different cell types. Extracellular vesicles transport bioactive molecules from one cell to an other leading to a biological response within the target cell. The aim of this project is to isolate and characterize microvesicles from the blood derivatives PRP and hypACTTM serum in order to determine their regenerative potential to fight osteoarthritis.
