Beschreibung
Sepsis stellt eine lebensbedrohliche Organdysfunktion als Folge einer schweren systemischen Entzündung dar, die durch eine Infektion ausgelöst wird. Weltweit werden etwa 31,5 Mio. Sepsisfälle jährlich registriert, und selbst bei bestmöglicher intensivmedizinischer Behandlung beträgt die Mortalität 15-25% bei Sepsis und bis zu 50% bei septischem Schock. Sepsis stellt eine der häufigsten und kostenintensivsten Erkrankungen im stationären Sektor dar. Die Verbesserung der Diagnostik, Therapie, Erfassung und Nachsorge von Sepsis wurde daher von der Weltgesundheitsorganisation WHO im Jahr 2017 zum prioritären Ziel erklärt. Dieses Projekt wird (i) zur Verbesserung der diagnostischen Möglichkeiten von Sepsis beitragen (raschere Diagnose für das Vorliegen einer systemischen Infektion), (ii) physiologisch relevante Zellkulturmodelle zur Charakterisierung der Rolle des Endothels in der Sepsis generieren, um neue Ansätze für die Behandlung von Sepsis zu finden; sowie (iii) den Effekt der Entfernung von Entzündungsmediatoren durch extrakorporale Adsorptionstherapien in septischen Patienten untersuchen. Das Projekt baut auf den im CD-Labor für Innovative Therapieansätze in der Sepsis entwickelten Methoden und gewonnenen Erkenntnissen auf. Dies gilt insbesondere für die Charakterisierung der Rolle extrazellulärer Vesikel in Inflammation und Sepsis sowie für Zellkulturmodelle zur Untersuchung der Aktivierung des Endothels unter septischen Bedingungen.
